Human Antibody Libraries

Dyax human antibody phage display technology allows for the rapid isolation of fully human, therapeutic target-specific antibodies from a human antibody library of billions of distinct antibodies.

We combine these libraries with in vitro selection strategies that are tailored to increase the yield of lead candidates with the desired therapeutic properties.

This technology offers reproducibility, high-throughput, and a level of control that is unmatched by other approaches. Our human antibody phage display libraries have resulted in multiple drug candidates in Phases 1, 2, and 3.

Dyax human antibody phage display libraries have been used to identify high-affinity, high specificity human antibodies for many distinct targets, including:

  • Cytokines
  • Cell surface receptors
  • Tumor cell markers
  • Viral antigens
  • Enzymes

Approach

Dyax's human antibody libraries are designed to facilitate the rapid production and purification of soluble Fabs and IgGs and, if needed, permits easy affinity maturation.

These proprietary libraries combine gene fragments from human donors with strategically designed synthetic diversity.

To achieve this, the library is designed with:

  • Human-donor-sourced fragments encoding:
    • Light-chain variable region
    • Heavy-chain CDR-3
  • Synthetic DNA encoding diversity in heavy-chain CDR-1
  • Synthetic DNA encoding diversity in heavy-chain CDR-2

These diverse gene products are displayed on phage as Fab antibody fragments.
